MALDI target for improved analysis of complex proteomics samples

چکیده
described. The new AnchorChipTM target provides substantial improvements to the LC-MALDI analysis of complex biological samples. The MTP AnchorChip 1536 TF can accommodate fractions collected from an extended LC gradient (up to four hours) or, alternatively, from multiple shorter LC gradients spotted in sequence. Complete MALDI-TOF/TOF analysis of all the spotted fractions can be easily achieved within a single run without the need of target exchange. This provides significant improvements regarding utilization of the high throughput capacity offered by high speed MALDI-TOF/TOF systems such as Bruker ́s ultrafleXtremeTM. Applying expanded LC gradients to the LC-MALDI analysis of complex proteomics samples leads to an improved depth of analysis resulting in a higher number of protein IDs and peptide assignments. In the benchmark example shown here, a single injection of 500 ng E.coli cell lysate digested with trypsin yielded more than 900 protein IDs based on more than 6000 assigned peptides.

The use of nLC-ESI-MS/MS in shotgun proteomics experiments and GeLC-MS/MS analysis is well accepted and routinely available in most proteomics laboratories. However, the same cannot be said for nLC-MALDI MS/MS, which has yet to experience such widespread acceptance, despite the fact that the MALDI technology offers several critical advantages over ESI. As an illustration, in an analysis of mode...
متن کاملProtein Stamping for Maldi Mass Spectrometry Using an Electrowetting-based Microfluidic Platform
MALDI-MS (matrix-assisted laser desorption/ionization mass spectrometry) is one of the most commonly used techniques for protein analysis. In conventional systems sample preparation is typically done in well-plates and transferred onto a MALDI target by robotic systems, which are complex, huge, expensive and slow. In this paper, we present a droplet-based microfluidic interface to transfer prot...

متن کاملA Nano LC-MALDI Mass Spectrometry Droplet Interface for the Analysis of Complex Protein Samples
The integration of matrix-assisted laser desorption ionization (MALDI) mass spectrometry with an upstream analytical separations (such as liquid chromatography and electrophoresis) has opened up new opportunities for the automated investigation of complex protein and peptide mixtures. The ability to efficiently analyze complex proteomic mixtures in this manner is primarily determined by the abi...
